Geldanamycin (GA)显著延迟和降低viperin的表达，表明IRF3在RAW264.7细胞中参与毒蛇蛋白的诱导 [2]。 Geldanamycin (GA) 是一种苯醌安沙霉素，可保护在培养的原代神经元中的氧葡萄糖剥夺 (OGD)/zVAD处理引起的神经元损伤。更重要的是，Geldanamycin (GA)以时间和浓度依赖性方式降低RIP1蛋白水平[3]。 Geldanamycin (GA)还会降低Hsp90蛋白水平，导致RIP1蛋白不稳定。这会导致RIP1蛋白的不稳定性，导致格列达霉素治疗后RIP1蛋白水平降低，但RIP1 mRNA水平没有降低[3]。 Geldanamycin (GA) 被确定为Hsp90的第一个天然产物抑制剂，与Hsp90的N末端ATPase结构域结合以抑制其伴侣功能，并通过凋亡机制显著诱导肿瘤细胞死亡 [4]。 |
